1) What rough-in depth does the American Standard Flash RU107E shower valve allow from the finished wall?
The Flash rough-in valve sets between 1-3/4 inches and 2-3/4 inches from the finished wall to the back of the valve. That one inch of range is what lets you use it behind tile, thin panel, or a thicker wall build without repositioning the cross brace, and the plaster guard doubles as the depth guide while you set it. On thin-wall work the plaster guard can also act as mounting support.

Either direction works. The valve body is symmetrical, so it can be rotated 180 degrees to take supplies from above or below. Hot and cold can also be reversed quickly for back-to-back bathrooms where the stack feeds two showers off one wall.
