Yes, that's the normal use. A 3/8 x 4 hex lag driven into wood framing or a plywood backboard is a standard anchor point for strut, hanger brackets, pull boxes and backboards, and the 250 piece box covers a full rough-in. Into concrete or block you'd need a concrete anchor or a lag shield instead, since a lag alone won't hold in masonry.
2) Do I need a nut and washer with the Metallics BLS25PLB?
No nut. A lag bolt threads directly into the wood, so nothing is needed on the back side, and nothing is supplied with it. A flat washer under the hex head is good practice so the head doesn't crush the surface as you torque it down, but washers are ordered separately.
