1) Is this red Sta-Kon terminal rated for 600 volt circuits?

2) Why choose a nylon insulated ring terminal over an uninsulated lug with heat shrink?
A nylon insulated terminal like the RL737 arrives with the sleeve already on the barrel, so you crimp and you are done, no shrink tube staged on the conductor first. Nylon also takes more abuse during pull-in than vinyl. If you need a specific jacket color coding scheme or a long sealed transition at the barrel, an uninsulated lug plus adhesive-lined shrink gives you that control instead.
3) Will a ring terminal work where I currently have a spade or fork terminal on a 3/8 inch stud?
Yes, and it is the better choice on anything that vibrates or carries real current. A ring tongue is captive, so the connection cannot slide off the stud if the nut loosens, whereas a fork can walk out. The tradeoff is that you have to remove the nut completely to install the RL737, so it is not a quick-disconnect point.
4) What crimp tool do I need for a 4/0 Sta-Kon ring terminal?
A 4/0 copper barrel is well past hand-tool range. It needs a hydraulic or heavy mechanical crimper with the die that matches the terminal barrel, and the crimp has to be indented per the terminal maker's die chart to hold the UL E9809 recognition. Pliers or a standard insulated-terminal plier will not form this barrel.
5) What does the red insulation on this Sta-Kon mean, and does it indicate the wire gauge?
The RL737 uses red nylon insulation, but on heavy lugs like a 4/0 the color is not the small-terminal AWG code (red/blue/yellow for 22-16, 16-14, 12-10). Treat the color as identification only and size off the stamped 4/0 and 3/8 ratings.
