Nut down means the slip nut on the tailpiece faces downward toward the trap, with the branch positioned to suit that layout. The branch sits 3 inches from the end, so measure from your strainer down to where your dishwasher or condensate hose comes in before ordering. If the drain line you need to tie in lands above that point, the nut down version puts the branch too low.
