1) Will the JDP3 anchor hold in drywall or hollow block?
No. The JDP3 is a drive pin anchor meant for solid base material, concrete, brick or solid masonry, where the body can expand against the hole wall. Drywall and hollow cells give it nothing to grip. For gypsum use a toggle or self drilling drywall anchor, and for hollow block use a toggle style anchor sized to the cell.
2) How many anchors come in a jar of Metallics JDP3?

3) Do I need a hammer drill and a setting tool, or can I install these by hand?

5) Are drive pin anchors removable if I have to take the fixture back off?
Treat them as permanent. A drive pin anchor expands when the pin is driven and does not back out like a screw, so removal normally means shearing or grinding the head and patching the hole. If the mount has to come down and go back up, use a threaded anchor or a machine screw anchor instead of the JDP3.
