Yes on the cross bore, but the trim prep is different. The 6243 STRETTO uses a 2-1/8 inch cross bore like any standard tubular set, and the backset is adjustable 2-3/8 or 2-3/4 inch. What differs is the face prep: it takes a narrow 1-1/4 inch door prep with 3-3/8 inch center-to-center hole spacing for the long 1-1/2 x 11-1/8 inch plate. A door drilled for a round rosette set will need the additional narrow-trim prep before this plate mounts.
2) Do I have to specify handing on the Emtek 6243 STRETTO set?
Handing is required, right hand or left hand, and it is a required selection on the order. Emtek calls out handing specifically for leversets, so if you are pairing this privacy trim with a Medici, Napoli, Octagon or Padua lever, get the hand right or the lever will point the wrong way.

Not keyed. The 6243 is a privacy/bed/bath function, so it locks from the inside with a thumbturn or push button and has an emergency release on the outside instead of a cylinder. Choosing a lever is what triggers the handing requirement; knobs are non-handed in use but the set is still ordered by hand.
